摘要
目的探讨晚期结直肠癌湿热内蕴证的中医证候实质,为证候客观化研究提供思路。方法:选择晚期结直肠癌湿热内蕴证患者3例,以非湿热内蕴证患者6例作为对照,采集外周血,采用Agilent表达谱芯片筛选湿热内蕴证的差异表达基因,并进行基因功能注释及富集度分析。结果:晚期结直肠癌湿热内蕴证患者与对照组相比共筛选出168条差异表达基因,其中上调的75条,下调的93条。上调的基因功能,明显的趋势是湿热内蕴证大部分症状与炎症反应和免疫应答的过度表达相关,约占到显著性富集的62.5%:下痢脓血与和肝素结合的基因表达下调相关;与碳水化合物结合的基因表达下调可能是导致是湿热内蕴证患者嗜食肥甘厚味的原因。结论:应用表达谱芯片筛选差异表达基因,可以从分子生物学水平对晚期结直肠癌湿热内蕴证的证候实质的探索提供新思路;湿热内蕴证与炎症反应和免疫应答的过度表达、与肝素及碳水化合物结合能力减弱相关。
Objective To explore the essence that syndrome of dampness-heatin molecular biology,these patients are suffering from advanced colorectal cancer patients.And to provide ideas for the objective study of the syndrome.Methods:We chosed 3 syndrome of dampness-heat patients who are suffering from advanced colorectal cancer as the test group, and 6 patients who are not syndrome of dampness-heat as the control group.We collected blood,extracted RNA,using the Agilent microarray screening of differentially expressed genes of deficiency syndrome of both liver and kidney yin, and gene functional annotation and enrichment analysis, Pathway analysis.Results:Advanced colorectal cancer patients with syndrome of dampness-heat compared with the control group were screened out of 168 differentially expressed genes, which raised the 75, down the 93. Translation and differentially expressed genes functional enrichment analysis,We found a clear upward trend in differences in gene function:mostsyndrome of dampness-heatpatients with symptoms of over-expression of inflammation and immune response related, accounting for the significant enrichment of 62.5%.Pus and heparin binding associated with reduced gene expression.Gene expression and carbohydrate binding reduction may be the cause of syndrome of dampness-heat patients like to eat greasy food causes.Conclusions: We can provide ideas for essence that syndrome of dampness-heat in molecular biology by screening differentially expressed gene.Syndrome of dampness-heatand overexpression of an immune response, in combination with heparin and related carbohydrates weakeningrelated.
