磷酸化HSP27在胰腺癌中的表达与患者临床预后的相关性

目的通过检测磷酸化HSP27（pHSP27）在胰腺癌组织中的表达，结合胰腺癌患者临床资料探讨pHSP27的表达与患者临床预后的相关性，并分析其临床意义。方法：通过免疫组化Envision二步法检测49例胰腺肿瘤患者手术切除石蜡标本中pHSP27蛋白的表达水平，采用western blot方法验证pHSP27在胰腺癌患者组织中的表达，结合胰腺癌患者临床资料，使用Kaplan—Meier法分析随访患者手术切除标本中pHSP27的表达与患者临床预后的相关性。结果：49例标本中，有42例pHSP27高表达，7例为低表达；而western blot的结果显示癌组织中的pHSP27蛋白的表达量要远高于癌旁组织和良性肿块组织（P〈0．05），且pHSP27在胰腺癌中的表达差异与年龄、性别、肿瘤大小无明显相关性，而与组织分化、临床分期、淋巴结转移、远处转移有明显相关性。Kaplan—Meier生存分析结果显示，pHSP27在胰腺癌组织中的高表达与胰腺癌患者预后良存在相关性（P=0．0051）。结论：pHSP27在胰腺癌组织中明显高表达，提示pHSP27在胰腺癌的肿瘤发生、发展中发挥着重要的作用。同时，pHSP27的高表达提示着胰腺癌患者不良预后。

Objective To investigate the expression of pHSP27 （phosphorylated heat shock protein 27） in pancreatic carcinoma,Combiningwith pancreatic carcinoma patients clinical data, we investigate the relationship between the expression of pHSP27 and its clinical prognosis,and then analyze its clinical significance. Methods：Immunohistochemistry of Envision method were used to detect the expression of pHSP27 in49 pancreatic carcinoma samples, and western blot method were also used to detect the expression of pHSP27 in pancreatic carcinoma sampletissues. Combining with the clinical data of pancreatic cancer patients, the method of Kaplan-Meier was used to analyze the relationshipbetween the expression of pHSP27 and the corresponding clinical prognosis of follow-up patients. Results：In 49 cases of paraffin specimens, 42cases of carcinoma tissues of pHSP27 reveals positive expression, 7 cases of adjacent to carcinoma tissues of pHSP27 reveals negativeexpression. Western blot results show that the expression of pHSP27 in cancer tissue is much higher than adjacent to carcinoma tissue andbenign tumor tissue （P〈0.05）, and the expression difference of pHSP27 in pancreatic cancer has no significant correlation with age, sex, tumorsize, but has significant correlation with tissue differentiation, clinical stage, lymph node metastasis and distant metastasis. Kaplan-Meiersurvival analysis shows that the high expression of pHSP27 in pancreatic cancer tissues and the prognosis of patients with pancreatic cancerhave good correlation （P=0.0051）. Conclusion：pHSP27 significantly high express in pancreatic cancer tissues. It suggests that pHSP27 may playan important role in pancreatic tumor in tumorigenesis and development. At the same time, the high expression of pHSP27 reveals a poorprognosis of pancreatic carcinoma patients.
