献浆员中呼吸道合胞病毒中和抗体效价分布及高效价血浆筛选 被引量:2

Distribution of neutralizing antibody titer against respiratory syncytial virus in plasma donors and screening of high titer plasma

摘要 目的分析献浆员中呼吸道合胞病毒(respiratory syncytial virus,RSV)中和抗体效价分布情况,筛选高效价RSV中和抗体血浆,用于制备特异性RSV免疫球蛋白。方法对600份血浆进行倍比稀释,采用微量中和试验法对血浆中的RSV中和抗体效价进行检测,选择合适稀释度作为大规模血浆检测稀释点。以单一稀释度对2 976份血浆的RSV中和抗体进行检测,并对高效价混合血浆进行检测和评价。结果 600份血浆中,血浆RSV抗体效价≥1∶64占65.5%,≥1∶180占18.3%,≥1∶256占11.2%。以1∶200稀释度对2 976份血浆进行筛选,共筛选出463份血浆,占15.5%。463份混合血浆的RSV中和抗体效价达到普通混合血浆的4倍。结论经本方法筛选出高效价的血浆,用于制备特异性RSV免疫球蛋白是可行的。 Objective To analyze the distribution of neutralizing antibody titer against respiratory syncytial virus(RSV)in plasma donors, and screen high titer plasma for preparation of specific RSV immunoglobulin. Methods A total of 600 plasma samples were diluted 2-fold serially, and determined for neutralizing antibody titer against RSV by micro-neutralization assay, based on which the dilution was optimized for screening of high titer ones from 2 976 plasma samples. The screened plasma samples were determined and evaluated. Results The titers of 18. 3% of 600 plasma samples were not less than 1 ∶ 180, while 11. 2% were not less than 1 ∶ 256,and 65.5% were not less than 1 ∶ 64. From the 2 976 samples each at a dilution of 1 ∶ 200, 463 with high titer neutralizing antibody against RSV were screened, which accounted for 15. 5%. The titer of pool of the 463 samples reached 4 times of the common mixed plasma. Conclusion The screened high titer plasma was suitable for preparation of specific RSV immunoglobulin.
