《金匮要略》泽泻汤超临界萃取物对膜迷路积水模型影响的研究 被引量:4

摘要 目的:研究泽泻汤超临界萃取物对膜迷路积水豚鼠模型的影响。方法:采用醋酸去氨加压素复制膜迷路积水模型,以豚鼠颞骨切片耳蜗蜗管面积和蜗管加前庭阶面积之和之比值以及耳蜗及肾的水通道蛋白-2的免疫组化表达为药效学的主要指标,研究泽泻汤超临界萃取物对膜迷路积水豚鼠模型的作用。结果:模型组与正常对照组比较有非常显著性差异(P<0.01);模型组与泽泻汤超临界萃取液治疗组比较有非常显著性差异(P<0.01)。水通道蛋白-2免疫组化表达实验组较模型组减弱。结论:泽泻汤超临界萃取物对膜迷路积水模型有较好作用,且机制可能为抑制其内耳及肾脏水通道蛋白-2的表达。 Objective :To study the effect of Alisma Tang' s supercritical extraction fluid on hydrolabyrinth guinea pig models. Methods ; The guinea pigs were intraperitoneally injected with Desmopressin to establish rat models of hydrolaby- rinth, the area of temporal bone slice cochlear duct/area of cochlear duct and scala vestibnli ratio and immunohistochemi- cal expression of water channel protein - 2 were chosen as indexes to study the effect of Alisma Tang~ supercritical extrac- tion fluid. Results :There was a significant difference between normal control group and model group (P 〈 0.01 ). There was a significant difference between Alisma Tang' s supercritical extraction fluid group and model group ( P 〈 0.01 ). And immunohistochemical expression of water channel protein - 2 of Alisma Tang' s supercritical extraction fluid group was weaker than the model group' s. Conclusion:The supercritical extraction fluid has the effect of reducing hydrolabyrinth and the mechanism may be to curb the inner ear and kidney water channel protein -2 expression.
