摘要 目的:研究蜣螂水提取物对小鼠的镇痛抗炎作用及其机制。方法:采用醋酸扭体法、甩尾法、热板法研究蜣螂水提取物的镇痛作用;采用二甲苯致小鼠耳肿胀实验、鸡蛋清致小鼠足趾肿胀实验、醋酸致小鼠腹腔毛细血管通透性增加实验观察蜣螂水提取物的抗炎作用;测定胸膜炎大鼠胸腔渗出液中总蛋白含量、血清中前列腺素E_2(PGE_2)与肿瘤坏死因子(TNF-α)含量,探讨其镇痛抗炎作用机制。结果:蜣螂水提取物低、中、高剂量均能显著延长小鼠在热板法和甩尾法中的耐热时长,减少醋酸所致的小鼠扭体次数,抑制二甲苯所致的小鼠耳肿胀、鸡蛋清所致的小鼠足趾肿胀以及醋酸所致的小鼠腹腔毛细血管通透性增加,并能有效降低胸膜炎大鼠胸腔渗出液中蛋白含量、血清中前列腺素E_2(PGE_2)与肿瘤坏死因子(TNF-α)含量。结论:蜣螂水提取物具有明显的镇痛抗炎作用,其作用与降低PGE_2和TNF-α含量有关。 Objective : To investigate the analgesic and anti - inflammatory effect and the action mechanism of water extract from dung beetle on mice. Methods: Using acetic acid- induced writhing method,tail flick test and hot plate test were used to confirm whether the water extract from dung beetle has the analgesic effect on mice. The anti - inflammatory experiments concluded ear - swollen in mice induced by xylene, toe swoIIen in mice induced by ovi albumin and permea- bility of the capillary wall increasing induced by acetic acid. Contents of protein, PGE2 and TNF - α in pleural effusion of pleuritic rats were determined to research the action mechanism. Results : The water extract from dung beetle in low, mid- dle and high doses can prolong the heat - resisting of mice in the hot plate test and tail flick test and reduce the writhing times of mice caused by acetic acid. Besides, they all can inhibit mice ear - swollen induced by xylene, mitigate toe swol- len induced by ovi albumin and suppress the permeability of the capillary wall increasing induced by acetic acid and re- duce the contents of protein in the extravasate, PGE2 and TNF - α in blood from rat pleurisy. Conclusion: The water extract from dung beetle has anti - inflammatory and analgesic effect which is related to the inhibition of the production of pro- tein, PGE2 and TNF - α.
