摘要
以慢生型大豆根瘤菌(B radyrhizobium japonicum)H W-05的基因组D N A作为模板,PC R扩增固氮酶基因nif D K上下游片段,融合后与p K18m obsac B载体相连。采用三亲本杂交的方法,将构建的载体转化到慢生大豆根瘤菌中,构建此工程菌株为研究大豆的固氮能力奠定了基础。
The slow-growing rhizobia(Bradyrhizobium japonicum) HW-05 genomic DNA was taken as a template,PCR amplification of nitrogenase gene nif DK downstream fragment;connected with the fusion of p K18 mobsac B carrier.Using three parental hybridization to transform the constructed vector into slow-growing rhizobia,and the project has built foundation of the research on nitrogen fixation ability of soybean.
