摘要 目的:探讨力竭游泳运动对小鼠空间学习记忆的影响,以及小鼠前脑皮层和海马胶质细胞源性神经营养因子(GDNF)的作用。方法:建立反复力竭游泳运动小鼠模型,通过Morris水迷宫检测小鼠空间学习记忆能力的变化,采用免疫组化检测力竭游泳运动后即刻(0h)、12h、24h和1w小鼠前脑皮层、海马胶质细胞源性神经营养因子(GDNF)的表达。结果:与对照组小鼠比较,反复力竭游泳组小鼠在力竭游泳后12h和24h小鼠的逃避潜伏期(训练周期1)显著增加,第一象限停留时间明显减少,小鼠空间学习记忆能力显著降低(P<0.01,P<0.05);与对照组小鼠比较,在力竭游泳后即刻(0h)和12h小鼠各脑区GDNF阳性细胞平均目标灰度值均明显增多(P<0.05,P<0.01),力竭游泳后24h,小鼠海马DG和前脑皮层GDNF阳性细胞平均目标灰度值明显增多(P<0.05);力竭游泳后一周,无显著性差异(P>0.05)。结论:力竭游泳运动导致了小鼠空间学习记忆功能减退,同时引起了海马和前脑皮层GDNF的表达出现不同程度的升高;GDNF参与了中枢疲劳产生与恢复的生物学调控过程。 Objective:To study the effect of exhausted swimming exercise on spatial learning- memory function,and the role of glial cell line-derived neurotrophic factor(GDNF)of prefrontal cortex(PFC)and hippocampus(HP)in mice.Methods:The repeatedly exhausted exercise model mice were applied.The spatial learning-memory ability of mice were tested using Morris water maze task.The expression of GDNF in HP and PFC were detected by immunohistochemical method,at 0(immediately),12 h,24hand 1week after repeatedly exhausted exercise,respectively.Results:Compared with control mice,the escape latency were significantly increased,and the swimming time of first quadrants were significantly decreased at 12 h,24hin exhaustive exercise group mice(P<0.01,P<0.05),after repeatedly exhausted exercise.The ability of spatial learning and memory of the exhaustive exercise group mice were significantly reduced.Compared with control group mice,the average target gray value of GDNF in PFC and HP of exhaustive exercise group mice were significantly increased at 0hand 12h(P<0.05,P<0.01),and were significantly increased at 24 hin DG and PFC of mice brain,after repeatedly exhausted exercise.There were not significant difference the average target gray value of GDNF at 1 week after exhausted exercise.Conclusion:Exhausted swimming exercise resulted in a decrease in spatial learning and memory in mice,and increased the expression of GDNF in HP and PFC;GDNF is involved in the biological control of central fatigue and recovery.
