前交叉韧带损伤后膝关节腔u-PA含量变化及意义 被引量:1

摘要 目的探讨前交叉韧带损伤后膝关节腔内u-PA变化情况及意义。方法 40只新西兰兔一期建立单侧前交叉韧带损伤模型。损伤术后2周、3周和4周时采用ELISA方法检测膝关节液中u-PA含量变化。损伤术后3周时采用HE染色和免疫组织化学检测关节腔内滑膜组织病理变化和u-PA表达情况。结果前交叉韧带损伤后膝关节腔内u-PA生成增加,术后3周时关节腔内u-PA含量明显高于假手术组(P<0.05)。术后3周时组织学检测显示滑膜组织病理变化明显,滑膜组织内u-PA广泛表达、阳性细胞数量增多(P<0.01)。结论前交叉韧带损伤后膝关节腔内u-PA含量增加,可能与滑膜组织病理改变有关。 Objective To investigate the levels change and its significance of urokinase-type plasminogen activator in articular cavity after anterior eruciate ligament injury. Methods Forty New Zealand rabbits were employed in this study and the transection of anterior crueiate ligament was created in the unilateral knee. The concentrations of urokinase-type plasminogen activator in joint fluid were detected by ELISA postoperatively at 2,3,and 4 weeks after injury. At 3 weeks after injury the pathological change and expression of urokinase-type plasminogen activator were further evaluated in the synovium. Results The level of urokinase-type plasminogen activator in joint fluid had dramatically changed. The levels of urokinase-type plasminogen activator were significantly elevated at 3 weeks after injury than that of sham operation group (P〈0.05). At 3 weeks after injury the pathological changes were proven significant in the synovium. Moreover,in the synovium the expression of urokinase-type plasminogen activator was elevated and the number of the positive cells markedly increased (P〈 0.01). Conclusion The levels of urokinase-type plasminogen activator in the knee joint are significantly increased which may be related to the pathological changes of the synovium.
