白芍总苷对EAT小鼠Th1/Th2型细胞因子表达的影响 被引量:20

摘要 目的:观察不同浓度白芍总苷(total glucosides of paeonia,TGP)对实验性自身免疫性甲状腺炎(experimental autoimmune thyroiditis,EAT)小鼠Th1型细胞因子(IFN-γ)和Th2型细胞因子(IL-4)表达的影响。方法:在建立小鼠EAT模型第3周起灌胃给药,给药组分别给予不同浓度TGP混悬液和雷公藤多苷片混悬液,模型对照组给予等量的0.5%羧甲基纤维素钠(CMC-Na)。用流式细胞仪分析各组小鼠脾细胞分泌IFN-γ和IL-4的T细胞比例。结果:雷公藤多苷和高剂量TGP可显著抑制EAT小鼠脾细胞IFN-γ和IL-4的过高表达,而中、低剂量TGP抑制作用较差。结论:TGP的治疗作用可能与调节EAT小鼠亢进的细胞免疫和体液免疫功能及平衡细胞因子表达有关。 AIM: To observe the expression of Th1 type cytokines( IFN-γ) and Th2 type cytokines( IL-4) on experimental autoimmune thyroiditis( EAT) mice by the different concentrations of total glucosides of Paeonia( TGP). METHODS: After establishment of model in the third week,each medicine group was given different concentrations of TGP suspension or solution of triptergium glycoside. The model group was treated by 0. 5% sodium carboxymethyl cellulose( CMC-Na) in the same volume.Those T cells secreting IFN-γ or IL-4 were analyzed by flow cytometry. RESULTS: Triptergium glycoside and high-dose TGP can apparently inhibit the excessive expression of IFN-γ and IL-4 in EAT mice splenocyte,while inhibitory effects of middle and low-dose TGP are weaker. CONCLUSION: TGP's therapeutic effect on EAT mice is related to the regulation of EAT mice's cell immune and humoral immune function and the expression of the cytokines.
