雌二醇、胰岛素对子宫内膜癌细胞增殖、凋亡及相关基因表达的影响 被引量:4

摘要 目的探讨胰岛素、雌二醇对ER阳性Ishikawa细胞和ER低表达HEC-1A子宫内膜癌细胞株的增殖、凋亡的影响及相关基因表达的影响。方法应用MTT法、流式细胞和荧光定量PCR技术分别检测胰岛素、雌二醇、胰岛素+雌二醇作用后Ishikawa和HEC-1A细胞的增殖、凋亡和VEGFA、b FGF、DKK1基因表达的情况。结果 10-8mol/L雌二醇、胰岛素对Ishikawa和HEC-1A细胞均具有不同程度的促增殖作用和协同性,也具有抑制Ishikawa、HEC-1A细胞凋亡的作用,对HEC-1A细胞的协同性更强;HEC-1A细胞VEGFA和b FGF mRNA协同促进表达作用显著(P<0.05),Ishikawa细胞的抑制因子-1(DKK1)表达则具有协同降低作用,差异有统计学意义(P<0.05)。结论雌激素、胰岛素对子宫内膜癌细胞具有促进增殖、抑制凋亡的作用,在不同处理条件下两者具有协同促进相关基因表达的作用。 Objective To study the effects of estradiol and insulin on human endometrial tumor cell line Ishikawa and HEC - 1A and to investigate the changes of proliferation, apoptosis and gene expression in these cells. Methods The variation of proliferation, apoptosis and gene expression (VEGFA,bFGF and DKK1 ) in estradiol were detected by using MTr,Flow cytometry and Fluorescence quantitative PCR technique,and insulin and both adding group between Ishikawa and HEC - 1A cell. Results The proliferation of both cell lines were promoted and reflected the character of cooperativity by 10-Smol/L estradiol and insulin stimulating. The apoptosis in both cell lines were inhibited and the HEC - 1A was found with higher cooperativity (P 〈 0. 05 ). The mRNA expression of VEGFA and bFGF in HEC - 1A cells were increased by the stimulus with estradiol and insulin group (P 〈 0.05 ). Whereas, these two genes were decreased in Ishikawa cells at the same stimulus ( P 〈 0.05 ). Conclusion Estradiol and insulin can affect the human endometrial tumor cell's proliferation and apoptosis and they have the character of cooperativity to improve the related gene expression to some degree.
