摘要
目的:观察麦纤散对UC大鼠结肠组织细胞外信号调节激酶1(extra cellular regulated protein kinases 1,ERK1)、细胞外信号调节激酶2(extra cellular regulated protein kinases 2,ERK2)mRNA表达的影响,探讨其作用机制。方法:将实验动物分为6组,采用三硝基苯磺酸(trinitro-benzene-sulfonic acid,TNBS)法造模,造模成功后,分别连续灌胃给药2周观察大鼠一般情况。末次给药24h后处死大鼠取结肠,运用RT-PCR法检测其ERK1、ERK2 mRNA表达水平。结果:麦纤散能明显改善UC大鼠的一般情况;与模型组相比,美沙拉嗪组、麦中组、麦高组大鼠结肠组织中ERK1 mRNA显著增加(P<0.05),美沙拉嗪组、麦中组ERK2 mRNA的表达亦显著增加(P<0.05)。结论:麦纤散可改善UC大鼠一般情况,其作用机制可能与上调结肠组织中ERK1、ERK2 mRNA相对表达量关联。
Objective:To observe the influence of maixiansan on expressions of ERK1 gene and ERK2 gene in colon of the rats with ul-cerative colitis ( UC) , and to identify its underlying mechanism. Methods:Experimental animals were divided into six groups and trea-ted with TNBS to induce ulcerative colitis. After successful modeling, each experimental group was administrated NS, mesalazine, and Maixiansan respectively with two weeks, and the general situation was observedand. The colon tissue was collected 24 hours later after the last administration, and the expressions of ERK1 gene and ERK2 gene in colon tissue was detected by means of RT-PCR. Results:Maixiansan can obviously improve the general situation of UC rats; compared with normal group, the expression of ERK1 mRNA was increased in mesalazine group, Maixiansan middle-dose group and Maixiansan high-dose group; the expression of ERK2 mRNA was also increased in mesalazine group and Maixiansan middle-dose group. Conclusion:The significant effect of Maixiansan on UC rats may be attributed to regulate the expression of ERK1 and ERK2 in colon.
