摘要
目的、构建用于人N末端前钠尿肽(NT-proBNP)高灵敏检测的电化学传感器。方法采用自组装方法将牛血清清蛋白-碳纳米管复合物(BSA-MWNTs)田栽在洁净的玻碳龟板表面,结合共价交联固定化技术和亲和素-生物素信号放大技术,制得NT-proBNP高灵敏免疫传感器,优化实验条件,采用电化学方法对电极的响应特性进行研究。结果在最优实验条件下,该传感器对NT-proBNP响应的线性范围为1-30ng/mL和30-150ng/mL,检出限为0.03ng/mL。结论该方法可实现对NT-proBNP的超灵敏检测。
Objective To construct an electrochemical immunosensor for detection NT-proBNP with high sensitivity. Methods The proposed immunosensor was prepared by the follows:firstly,the hybrid material BSA- MWNTs was self-assembled onto the surface of cleaned glass carbon electrode;subsequently,streptavidin was covalently linked to the modified electrode;then, biotin labeled antibody could be absorbed to the electrode by the reaction between streptavidin and biotin. The effects of experimental paramenters have been investigated. Results Under the optimal experiment conditions,the prepared immunosensor displayed a wide liner relationship between 1- 30 ng/mL and 30-150.0 ng/mL with a detection limit of 0.03 ng/mL. Conclusion The NT- proBNP imunosensor can be utilized for the detection of NT-proBNP with low concentration.
