摘要
脱嘌呤/脱氧嘧啶核酸内切酶1(APE1),又称为氧化还原因子1(Ref-1),是一种具有双功能酶活性的生物大分子,不仅具有核酸内切酶活性,能修复DNA的损伤,而且具有氧化还原功能,可通过调控多种转录因子的转录活性实现对基因表达的干预。研究发现APE1/Ref-1在卵巢癌中呈高表达状态,并与肿瘤的发生、发展及预后有关。APE1/Ref-1因其与卵巢癌的密切关系,而成为极具潜力的肿瘤基因治疗的新靶点。该文就APE1/Ref-1的定位、分子结构、生物学功能及其在卵巢癌发生发展研究方面的进展作了一系列的回顾与总结。
Apurinic/apuriminidic endonuclease 1( APE1),also named redox factor-1( Ref-1),is a biomacromolecule with bifunctional enzyme. APE1 not only has the endonuclease activity to repair DNA injury,but also has a redox function,which interferes the gene expression by regulating the activity of a variety of important transcription factors. Recent studies have suggested that the expression of APE1 / Ref-1 in ovarian cancer is high and it is correlated with tumor occurrence,progression and prognosis. Thus,APE1 / Ref-1 becomes a new target for cancer gene therapy because of its correlation with ovarian cancer. In this paper,we made a series of review and summary concerning the allocation,structure and functions of APE1 / Ref-1 as well as the research progress in ovarian cancer.
