摘要 目的:建立四味雪莲花颗粒中大黄酚、大黄素的含量测定的方法。方法:采用反相高效液相色谱法(reversed phase high-performance liquid chromatography,RP-HPLC)测定四味雪莲花颗粒中大黄酚、大黄素的含量,采用Hypersil-ODS 2(250 mm×4.6 mm,5μm)色谱柱,以甲醇-(体积分数)0.1%磷酸溶液(8:2)作为流动相,以254 nm作为检测波长,以1m L·min^(-1)作为流速。结果:根据色谱图,大黄酚进样量为0.004 52~0.045 2μg,该范围与峰面积呈明显的线性关系(r=0.998,P<0.05),平均回收率为99.15%,RSD为0.85%。大黄素进样量为0.003 19~0.031 90μg,该范围与峰面积呈明显的线性关系(r=0.997,P<0.05),平均回收率为99.78%,RSD为1.13%。结论:反相高效液相色谱法用于测定四味雪莲花颗粒中大黄酚、大黄素的含量中具有高度灵敏度、准确度与简便性,有助于控制四味雪莲花颗粒的质量。文献引用:李奎,贾少谦.四味雪莲花颗粒中大黄酚、大黄素含量测定方法[J].中医学报,2016,31(8):1153^(-1)155. Objective:To establish content determination method of chrysophanol and rheum emodin in Siweixuelianhua granule. Methods : Content determination of chrysophanol and rheum emodin in Siweixuelianhua granule were used by reversed-phase high performance liquid chromatography (RP-HPLC) method,Hypersil ODS-2 (250 mm ×4.6 mm,5μm) chromatographic column, methanol -0.1% phosphoric acid solution (8:2) as mobile phase,254 nm was detection wavelength, and 1 mL· min^-1 as velocity. Results:According to the chromatogram, rhubarb phenol sample quantity was 0.004 52 -0.045 2 μg,the scope and peak area was showed obvious linear relationship ( r = 0.998, P 〈 0.05 ), the average recovery rate was 99.15 %, RSD was 0.85 %. Emodin sample quantity was 0. 003 19 - 0. 031 90 μg, the scope and peak area was showed obvious linear relationship ( r = 0. 997, P 〈 0.05 ) ,the average recovery rate was 99.78% , RSD was 1.13%. Conclusion:RP-HPLC has highly sensitivity, accuracy and sim- plicity for the content determination of chrysophanol and emodin in Siweixuelianhua granule, it helps to control the quality of Siweixuelianhua granule.
