摘要 建立了免疫亲和柱净化一超高效液相色谱法测定花生酱中黄曲霉毒素B1、B2、G1、G2的方法。试样经甲醇一水(55+45)提取,振荡、离心后,下层提取液经过滤、稀释、玻璃纤维滤纸二次过滤后,通过含有黄曲霉毒素特异性抗体的免疫亲和柱,以甲醇洗脱毒素。采用超高效液相色谱仪结合荧光检测器进行分析。结果表明,方法的检出限为0.1μg/kg,定量下限为1.0μg/kg,黄曲霉毒素Bl的回收率为88%~102%;黄曲霉毒素B1的重现性为2.5%~6.5%。该方法操作简便、准确、回收率高、重现性好,可用于花生酱中黄曲霉毒素B1、B2、G1、G2的检测。 A method for the determination of aflatoxins in edible oil was established. Samples were extracted with methanol - water (55 + 45 ) solution. After oscillated and centrifugated, the lower layer of extraction was filtered, diluted with water and filtered through glass microfiber filter paper. The filtrate was then passed through immunoaffinity column, and the toxins were eluted with methanol,then subjected to RPLC /FLD analysis. The LOD of this method is 0. 1 μg/kg and the LOQ is 1.0 μg/kg. The reeoveries of aflatoxin B1 ranged from 88 % to 102%, with relative standard deviations ( RSDs ) of 2. 5 % -6.5 %. The method is simple, rapid and aecurate, and is applicable for the determination of af/atoxins in peanut butter.
