秀珍菇子实体多糖PFP-m的膜法分离及其性质研究

摘要 以秀珍菇子实体为原料,经闪式提取、Sevag法脱蛋白、不同孔径的滤膜分离,得到秀珍菇子实体多糖(PFP-m)。采用光谱分析和比色等方法对其部分结构和性质进行测定。结果表明:PFP-m为均一组分,分子质量为3.27×10~4Da;具有多糖的特征吸收峰;PMP衍生化结合HPLC法得出其单糖组成及摩尔比为甘露糖∶鼠李糖∶葡萄糖∶木糖∶阿拉伯糖=3.10∶1.00∶22.99∶6.85∶4.59。PFP-m具有多糖的一般理化性质,总糖含量为93.16%。与柱层析法相比,膜超滤法操作简单,效率高,适合秀珍菇子实体多糖PFP-m的分离纯化。 A polysaccharide( PFP-m) was obtained from the fruit body of Pleurotus geesteranus by a sequential processing of flash extraction,deproteinization and separation through different pore ultrafiltration membrane. Its partial properties and structure were studied by spectrographic method and colorimetry. The results showed that PFP-m was a homogeneous polysaccharide. Its Mw was 3. 27 × 10~4 Da. Typical polysaccharide absorption of PFP-m was in its IR spectrum. PFP-m consists of Man,Rha,Glc,Xyl,Ara,in molar ratios of 3. 10 ∶ 1. 00 ∶ 22. 99 ∶ 6. 85 ∶ 4. 59 by PMP-HPLC. PFP-m has general physical and chemical properties of polysaccharide and its total sugar content were93. 16 %. Compared with column chromatography,ultrafiltration membrane method was easier to operate and more efficient. It was suitable for separation and purification of polysaccharide PFP-m from pleurotus geesteranus fruit body.
