摘要
目的观察养心2号方对慢性心力衰竭(CHF)大鼠血清炎症因子肿瘤坏死因子-α(TNF-α)、白介素-10(IL-10)及对诱导型一氧化氮合酶(iNOS)的影响,探讨其治疗CHF的作用机制。方法将45只SD大鼠随机分为正常对照组(Con组,n=6)及CHF组(n=39)。CHF组采用连续皮下注射异丙肾上腺素28d建立大鼠慢性心力衰竭模型。造模成功后,将存活的CHF组大鼠随机分为模型组(M组)、卡维地洛组(C组)及养心2号组(Y组),分别灌胃蒸馏水、卡维地洛及养心2号方;Con组大鼠灌胃蒸馏水,连续灌胃28d后,分别检测各组大鼠血清TNF-α、IL-10浓度及iNOS活力。结果与Con组比较,M组TNF-α浓度增高,IL-10浓度下降,iNOS活力增高,差异有统计学意义(P<0.01);与M组比较,各用药组大鼠血清TNF-α浓度下降、IL-10浓度增高,iNOS活力下降,差异有统计学意义(P<0.05)。结论养心2号方可降低CHF大鼠TNF-α水平,升高IL-10水平,降低iNOS活力,从而保护心肌、改善心功能,这可能是其治疗CHF的作用机制。
Objective To study the effect of Yangxin 2 decoction ( YXD 2)on the levels of tumor necrosis factor alpha ( TNF α)and interleukin 10( IL 10) and induced nitric oxide synthase( iNOS) in rats with chronic heart failure( CHF) .To explore the mecha -nism of YXD 2 for the treatment of CHF.Methods Forty five male Sprague Dawley ( SD)rats were divided into normal group( Con group, n =6)and CHF group( n =39) .The model group was injected hypodermic with ISO for 28 consecutive days .After themodel is successfully established in CHF, the survival rats were randomly devided into three groups: model group ( group M) , Carve -dilol group ( group C)and YXD 2 group( group Y) .Group C and group Y were received gastric gavage of carvedilol and YXD 2, re -spectively.The Con group and group M were received gastric gavage of distilled water.After 28 days of dosing, the levels of serumTNF αand IL 10 and iNOS were tested,Results Compared with the Con group, the levels of serum TNF αwere increased ingroup M( P 〈0.05) , the levels of serum IL 10 were decreased in group M( P 〈0.05) , and iNOS were increased in group M.Com -pared with group M, the levels of serum TNF αwere decreased, the levels of serum IL 10 were increased and iNOS were de -creasedin group C and group Y ( P 〈0.05) .Conclusion YXD 2 can reduce the concentration of serum TNF αand increase theconcentration of serum IL 10, reduce the concentration of serum iNOS, and improve cardiac function.
