摘要
目的探讨丹皮酚预处理对脑缺血再灌注(CI/R)损伤大鼠脑组织中核因子-κB(NF-κB)的影响。方法采用四血管阻塞10min再灌注12h方法建立CI/R损伤大鼠模型,分为模型组、丹皮酚不同剂量组(分别为5mg/kg、10mg/kg、20mg/kg)、假手术组,术前7d开始腹腔注射给药,每日1次,术后1h给药1次。再灌注12h后,免疫组化法检测各组脑组织海马CA1区NF-κB的水平。结果丹皮酚高剂量、中剂量可降低神经行为学评分,与模型组评分比较差异均有统计学意义(P<0.05)。丹皮酚不同剂量均可降低脑组织NF-κB表达,与模型组比较差异均有统计学意义(P<0.05)。结论丹皮酚预处理对CI/R损伤大鼠的脑保护作用可能与其下调NF-κB的表达、减轻脑组织的炎症反应等有关。
Objective To investigate the effect of paeonol pretreatment on nuclear factor κB ( NF κB)in rats with cerebral ischemia/reperfusion ( CI/R)injury.Methods Rat model with CI/R was prepared by four vessel occlusion for 10 min followed by 12 hourreperfusion.Rats were then randomly divided into 5 groups ( n =10) sham, model, paeonol high, medium and low dose groups( 20mg/kg, 10 mg/kg and 5 mg/kg, respectively) .Paeonol was injected intraperitoneally once a day for 7 days before the operation andonce 1 h after the operation,respectively.The NF κB activities in brain tissue of CI/R gerbils were detected by immunohistochemis -try after 12 hour reperfusion.Resluts Paeonol ( 10 mg/kg and 20 mg/kg)evidently improved the neurological behavior scores in ratswith CI/R injury,compared with that of model group ( P 〈0.05) .The IOD values of NF κB in paeonol groups ( 20 mg/kg, 10 mg/kgand 5 mg/kg)were significantly lower than that of model group ( P 〈0.05) .Conclusion The effectiveness of paeonol pretreatment inrats with CI/R injury appears to be associated with the inhibition of NF κB in brain tissue.
