摘要
目的:观察天然植物冬凌草提取物冬凌草甲素对胃癌HGC-27细胞侵袭能力的作用,并分析其可能的作用机制。方法:体外培养HGC-27细胞,分别加入0、20、40、80μmol/L浓度的冬凌草甲素作用24h,Transwell小室法检测细胞侵袭能力的改变;ELISA法检测细胞上清液中PGE2的含量;采用Western-blotting检测环氧合酶-2(COX-2)、基质金属蛋白酶-9(MMP-9)的表达。结果:冬凌草甲素处理后,HGC-27细胞侵袭能力降低,MMP-9表达下调;且COX-2的表达及PGE2合成降低。结论:冬凌草甲素可能通过抑制COX-2表达、PGE2合成、降低MMP-9表达降低HGC-27细胞侵袭能力。
Objective:To explore the effect of oridonin on the cell invasiveness in HGC-27 gastric cancer cells and to investigate the mechanisms.Method:The experiments were divided into two groups,including control group and drug treated group.The control group received only the culture medium.While in drug treated group,after treated by oridonin in different concentrations(20,40 and 80μmol/L)respectively for 24 h,the invasion of HGC-27 cells was tested by Transwell assay.The expression of PGE2 in the culture medium was detected by ELISA.Western-blotting was used for COX-2and MMP-9protein analysis.Result:Compared with control group,the invasion of HGC-27 cells was significantly decreased in oridonin treatment group.Oridonin inhibited the PGE2 synthesis.And the data of western blot showed that oridonin decreased the expressions of COX-2and MMP-9.Conclusion:Oridonin inhibits the cell invasion in HGC-27 cells,and the effect may be associated with inhibition of the expression of COX-2and PGE2 synthesis,thus down-regulating the expression of MMP-9.
