江苏沿海滩涂植物根际促生菌的筛选鉴定及其耐盐促生特性 被引量:11

Screening and identification of plant-growth-promoting Rhizobacteria from coastal tidal flats in Jiangsu province and its characterization of growth-promoting under salt stress

摘要 以1-氨基环丙烷-1-羧酸(ACC)为唯一氮源,从江苏省沿海滩涂植物根际土壤中筛选到4株促生菌株,测定其ACC脱氨酶比活力、产吲哚乙酸(IAA)和产铁载体能力。结果表明:菌株SC-12在以ACC为唯一氮源条件下,其ACC脱氨酶比活力达0.087U·mg-1,且具有较强的产铁载体能力。通过菌株形态、生理生化特征和16SrDNA序列分析初步鉴定该菌为同温层芽孢杆菌(Bacillus stratosphericus)。种子发芽试验表明,菌株SC-12在100、200 mmol·L-1硝酸盐胁迫条件下,能显著促进辣椒、番茄发芽率和芽长生长。 Four strains of plant growth-promoting rhizobacteria(PGGR) were isolated from plant rhizosphere soil in coastal tidal flat of Jiangsu province, based upon their ablility to utilize 1-aminocyclopropane-l-carboxylic acid (ACC) as the sole nitrogen source. Their ACC deaminase activities, properties of indole acetic acid (IAA) and production of sid- erophore were measured. The results indicated that the ACC deaminase activity of the strain SC-12 was 0. 087 U·mg-1 in mininmal medium containing ACC as the sole nitrogen source. In addition, the strain SC-12 also had strong sid- erophore-producing ability. Using morphology, physiological biochemical characteristics and 16S rDNA analysis, strain SC-12 was identified as Bacillus stratosphericus. Seed germination tests showed that the strain SC-12 could significantly improve seed germination and bud growth of pepper and tomato under the treatments of 100 mmol·L-1 and 200 mmol · L-1 NaNO3.
