摘要
采用响应面法对复合蛋白酶水解羊骨粉的工艺进行优化,建立了水解度与四个因素之间的数学模型,从而确定了pH 7.5时的最佳水解工艺:底物质量浓度为90 g/L、水解温度为55℃、水解时间为300 min、加酶量为5%(E/S),在此条件下,实际水解度为28.54%,与模型预测值接近。水解度越大,钙离子含量越高,二者相关系数达到0.909,在0.01水平极显著相关。在上述最优条件下水解100 g骨粉可生成游离钙离子35.19 mg。水解物中富含羊骨胶原肽和游离钙离子,提高了羊骨粉的利用价值,其生物学活性有待进一步研究。
Hydrolyzing conditions of sheep bone powder by Protemex were optimized via response surface design and analysis. The model of the degree of hydrolysis (DH) in response to 4 affecting factors was established. Hence, the optimal technique for hydrolyzing was determined that bone powder was 90 g/L, reacting at 55 ℃ for 5 hours, enzyme 5% (E/S), and the resulted DH reached to 28.54% which was quite close to the deduced theoretical value of the model. The content of Ca^2+ increased with the increased DH. The correlation coefficient reached to 0.909, the DH and Ca2+ content significantly correlated at the level ofp (0.01). Under the above optimal conditions, 35.19 mg Ca^2+ could be released from 100 g bone powder. The hydrolysate contains sheep ossein polypeptides and free ionic Ca^2+, therefore, the bio-availability of the sheep bone powder could be improved and the bioactivity of the hydrolysates needs further investigation.
