摘要
建立了一种高效液相色谱测定含乳饮料中γ-氨基丁酸(GABA)含量的方法。样品经乙腈-水沉淀后,上清液采用C18色谱柱分离,在338 nm波长下检测。γ-氨基丁酸在1.0-1 000μg/mL范围内线性关系良好,相关系数为0.999 8,以3倍、10倍信噪比计算方法的检出限、定量限,分别为0.47μg/mL,4.31 mg/kg。在40,50和60 mg/100 g的添加水平下,GABA的平均回收率为94.6%-104.4%,RSD为1.23%-3.05%。应用该方法检测了4种市售其他类食品中的GABA含量,结果表明该方法同样适用。
A new high performance liquid chromatography (HPLC) method for determination γ-aminobutyric acid in milk beverages was developed by automatic pre-column derivation. Milk beverages were precipitated by acetonitrile-water. The supematants were determined with C18 column and detected at 338 nm. The results showed that the linear range was 1.0- 1 000 μg/mL (r=0.999 8), and the limits of detection and quantitation were 0.47 lag/mL and 4.31 mg/kg, respectively. When adding levels were 40, 50 and 60 mg/100 g, the mean recoveries of γ-aminobutyric acid were 94.6%-104.4% with the relative standard deviations of 1.23% -3.05%. The method was successfully applied to the analysis of γ-aminobutyric acid in 4 other foods containing γ-aminobutyric acid.
