摘要
目的:探讨表皮生长因子受体(EGFR)抑制剂在舌鳞癌多药耐药中的作用。方法:免疫细胞化学技术和Western blot方法检测舌癌细胞系Tca8113及多药耐药细胞系Tca8113/CBP EGFR的表达,Western blot检测EGFR受到抑制后舌癌多药耐药细胞Tca8113/CBP耐药蛋白MRP,GTS-π的表达。结果:细胞系Tca8113和Tca8113/CBP均有EGFR的表达(P>0.05),EGFR受到抑制后细胞Tca8113/CBP的多药耐药蛋白MRP,GST-π的表达均下降(P<0.05)。结论:抑制EGFR可下调舌癌多药耐药细胞耐药蛋白的表达。
Objective:To study the effect of anti-EGFR agents on multidrug resistance( MDR) in tongue squamous cell carcinoma drug-resistance cell line Tca8113/CBP.Methods:Immunohistochemical staining and western blot assay were applied to detect EGFR expression in tongue squamous cell carcinoma cell line Tca8113 and its MDR cell line Tca8113/CBP, the protein expression levels of MRP, GST-πin cell line Tca8113/CBP which EGFR expression were inhibited were detected by western blot assay.Results:We found higher expression of EGFR in cell lineTca8113 and its MDR cell line Tca8113/CBP(P〉0.05).After the EGFR expression were inhibited,the expression MRP and GTS-πwere significant decrease in MDR cell line Tca8113/CBP( P〈0.05) . Conclusion:The anti--EGFR agents can lead to a downregulation of resistance protein expression.
