摘要
为探明沙田柚自交不亲和分子机理,以沙田柚(Citrus grandis var.Shatianyu Hort)自交和异交花柱为材料,对其进行转录组测序;通过差异分析获得相关基因,并研究其理化性质。结果表明:获得的Hsp90基因全长2 602bp(GenBank登录号:KU517851),开放阅读框(ORF)全长为2 100bp,编码699个氨基酸,编码的蛋白质分子量为80.55KDa,理论等电点为5.01,含有1个与HSP90超家族(HSP90superfamily)相同的保守结构域。沙田柚Hsp90蛋白氨基酸的同源性与甜橙(XM_006490568)、克莱门柚(XM_006421973)亲缘关系较近,同源性高达99%,属同一进化分支。
The self-pollinated and cross-pollinated style of C.grandis var.Shatianyu were used as test materials and based on the successful transcriptome sequencing technology,the authors identified a Hsp90 gene to explore the molecular mechanism of self-incompatibility.Results:Hsp90 gene was 2602 bp(GenBank accession number:KU517851)in length with an open reading frame(ORF)of 2100 bp,encoding 699 amino acids with deduced molecular weight of 80.55 KDa,and theoretical pI value of 5.01,and contained a HSP90 superfamily conserved domain.The homology analysis indicated that the Hsp90 protein shared 99% homology with that of Citrus sinensis(XM_006490568)and Citrus clementina(XM_006421973).Phylogenetic analysis revealed that Hsp90 gene showed closer kinship with that of C.sinensis and C.clementina,indicating that they belong to the same evolutionary branch.
