摘要
为获得羊耳菊种子生活力的最佳测定条件,采用四唑染色法,以氯化三苯基四氮唑(TTC)浓度、染色温度和染色时间为指标,通过正交试验和响应面试验对测定条件进行优化。结果表明:TTC浓度1.5%、染色温度40℃、染色时间3h为羊耳菊种子生活力测定的最佳条件,3个检测指标对种子生活力测定的影响为染色温度>染色时间>TTC浓度。
In order to find out the best determination conditions of seed viability of I.cappa,the method of 2,3,5-triphenyl tetrazolium chloride(TTC) was employed with indicators of TTC concentration,dyeing temperature and dyeing time,and the determination conditions were optimized by using orthogonal method and response surface methodology.Results:1.5%,40℃,3h were the best dyeing conditions of TTC concentration,dyeing temperature and dyeing time respectively.And the order of the three factors influencing seed viability test is dyeing temperature〉dyeing time〉TTC concentration.
