样品溶剂对高效液相色谱分析结果的影响

Effect of Sample Solvent on the Analytical Results of High Performance Liquid Chromatography

结合日常样品监测工作,针对相同条件下,以乙腈作为流动相,对三种常用溶剂下的苯并[α]芘进行了对比试验,实验表明:三种常用溶剂的洗脱能力由强到弱为二氯甲烷、乙腈、甲醇。由于溶剂极性的不同对样品的保留时间以及响应值产生了影响。在高效液相色谱方法的开发和应用中,应尽量使用与流动相比例相同或者相近的溶剂溶解样品,以消除影响。

Combining daily sample monitoring, under the same conditions, the benzene [ α ] pyrene of three kinds of solvents was compared with acetonitrile as mobile phase. Experimental results showed that the elution ability of three kinds of common solvents from strong to weak was dichloromethane, acetonitrile and methanol. The retention time and the response value of the samples were affected by different solvent polarity. In the development and application of chromatographic methods, the samples should be dissolved with mobile phase or similar solvent as could as possible, so as to eliminate the effect.
