摘要
[目的]对影响百蕊草愈伤组织培养的因素进行研究。[方法]以百蕊草幼嫩枝条和叶片为外植体,研究不同浓度6-BA、NAA、2,4-D或其组合,碳源以及pH等因素对百蕊草愈伤组织生长的影响。[结果]3种植物生长调节剂和碳源对愈伤组织的生长都有促进作用,对愈伤组织诱导生长的培养基最佳组合为MS+6-BA 1 mg/L+NAA 0.15 mg/L+2,4-D 0.1 mg/L,碳源中蔗糖显著促进了百蕊草愈伤组织的增长,以30 g/L的蔗糖为最佳;pH 5.8~6.0有利于愈伤组织的诱导和生长。[结论]该研究为后续研究百蕊草细胞悬浮培养体系的建立提供材料。
[Objective]The aim was tostudy factors influencing callus culture of Thesium chinense.[Method] Taking tender branches and leav-es of T.chinense as explants, effects of different concentrations of 6-BA, NAA, 2,4-Dand their combinations,carbon soueces as well as pH on callus were investigatedby using the orthogonal experiments.[ Result] The results showed that fitting concentration of plant growth regulators and carbon sources could all promote the growth of callus,the best medium for induction of callus is MS +mediumsupplemented with 6-BA 1 mg/L+NAA 0.15 mg/L+2,4-D 0.1 mg/L, in the carbon sources,sucrose markedly increased callus, the adding of 30 m/L sucrose was the best choice.The pH value was 5.8-6.0.[Conclusion] The study can provide materials for study on T.chinense cell suspension culture.
