磷酸化P38MAPK在退变的腰椎间盘髓核中的表达 被引量:4

THE PRELIMINARY STUDY ON THE EXPRESSION OF P-P38MAPK IN NUCLEUS PULPOSUS OF INTERVERTEBRAL DISC DEGENERATION

摘要 目的通过检测退变腰椎间盘组织中磷酸化P38MAPK的表达,探讨磷酸化P38MAPK在椎间盘退变过程中可能发挥的作用。方法选取2014年1月—2015年12月手术切除的腰椎间盘突出症患者的椎间盘组织,病理证实为退变腰椎间盘组织(退变组)50例;同时选取10例因外伤手术切除椎间盘的正常椎间盘组织(对照组)。采用免疫组织化学方法和ELISA方法对二组标本中p-P38MAPK的表达进行定位、半定量和定量分析,探讨p-P38MAPK与腰椎间盘退变的关系和意义。结果免疫组织化学结果显示阳性表达为腰椎间盘髓核细胞浆内可见棕黄色颗粒,p-P38MAPK的表达量退变组明显高于对照组,P<0.05。ELISA方法定量分析退变组椎间盘组织p-P38MAPK的含量明显高于对照组,P<0.05。结论在腰椎间盘退变过程中,p-P38MAPK具有广泛而复杂的生物学作用,考虑p-P38MAPK可能与炎症、应激反应的调控密切相关,其参与了TGF-β1等炎性应激因子的作用过程,认为是腰椎间盘退变的重要原因之一。 Objectives To investigate the expression of P38 MAPK and the significance in the disc degeneration tissues,and to clarify the roles in human intervertebral disc degeneration.Methods Fifty cases of the degenerative disc tissue were selected as the research samples(lesion group)which lumbar disc herniation surgical resection were performed from January 2014 to December 2015.At the same time 10 cases of trauma were as control group which received normal disc tissue surgical resection.The immunohistochemistry and ELISA specimens were used to detecte P38 MAPK in content between the two groups.Results Immunohistochemical results showed the expression of P38 MAPK in lesions group was significantly higher than that in control group,the positive was that brown granules can be seen in cytoplasm,the OD value in lesions group were significantly higher,P〈0.05.ELISA results showed the content of P38 MAPK in lesions group was significantly higher,P〈0.05.Conclusions In the disc degeneration,P38 MAPK are important mitogens activated protein kinase,thus plays its key role in disc degeneration.It has a broad and complex biological effects.We conside P38 MAPK involved inflammation,stress response is closely related to the inflammatory stress factor.The inflammatory factor is one of the important reason of disc degeneration.
