牡丹试管苗生根过程中酚酸类物质变化差异的研究 被引量:1

摘要 为探讨酚酸类物质与牡丹试管苗生根的关系,以生根率不同的牡丹品种‘太平红’、‘乌龙捧盛’和‘凤丹白’为材料,利用液相色谱-质谱联用(LC-MS)方法对试管苗生根过程中6种酚类物质成分定性定量分析,研究其在牡丹试管苗生根过程中的动态变化规律。结果表明:试管苗在生根过程中,3个品种体内的6种酚酸含量均发生了动态性变化,但不同品种变化规律不同。生根率越高的品种,其体内的芍药苷、苯甲酸、4-羟基苯甲酸含量越高,而没食子酸甲酯和莽草酸含量越低,没食子酸含量在3个品种中未能表现出类似的规律。本研究结果认为芍药苷、苯甲酸、4-羟基苯甲酸、没食子酸甲酯、莽草酸5种酚酸可能与牡丹试管苗生根密切相关,而没食子酸可能与牡丹试管苗生根关系不大。 The dynamic changes of six phenolic acids contents were evaluated by using high performance liquid chromatography-mass spectrometry during vitro rooting process of three cultivars ('Taipinghong', 'Wulongpengsheng', 'Fengdanbai') in order to study the relationship between phenolic acids and the occurrence of adventitious roots in Paeonia suffruticosa. Results showed that the six phenolic acids contents varied with time in different amplitudes during the vitro rooting process. However, there were differences in the dynamic changes among the three tree peony cuhivars. Paeoniflorin, benzoic acid and 4-hydroxy benzoic acid contents were higher in easy-to-root cultivar, while methyl gallate and shikimic acid contents were lower. However, no clear conclusion could be drawn for gallic acid content in correlation with the rooting ability of the three tree peony cuhivars. The results held that paeoniflorin, benzoic acid, 4-hydroxy benzoic acid, methyl gallate and shikimie acid were closely related to the rooting of tree peony cuhivars, the influence of gallic acid on rooting was little.
