壳聚糖降解酶的筛选及其HPLC分析 被引量:1

摘要 对木聚糖酶等四种酶水解壳聚糖的活性进行比较,结果表明:木聚糖酶对壳聚糖的水解活性最高;经木聚糖酶水解6 h后的壳聚糖的分子量集中在8.6 k Da左右,较其他酶解产物分子量小;对木聚糖酶组分进行分离纯化,纯化后木聚糖酶酶活损失5 200 IU,壳聚糖酶酶活损失300 IU;对酶组分HPLC分析表明,木聚糖酶中具有降解壳聚糖活性的组分分子量集中在34~45 kDa之间。 The activity of hydrolysing chitosan with the xylanase and the others enzymes was compared. The result indicates that the xylanase had highest activity to hydrolysing chitosan, hydrolysate's molecular weight distributed on 8.6 kDa which hydrolysed by xylanase after 6 h, the others had lower activity than xylanase. In the process of isolation and purification, the xylanase activity losed 5 200 IU, chitosanase activity losed 300 IU. Analysing the xylanase's component by HPLC, the results indicated that the xylanase consistsed of these molecular weight component was in the range of 34 and 45 kDa.
