摘要
目的检测PGRN在小鼠海马神经元HT22细胞中的表达水平,为进一步探讨颗粒蛋白前体(PGRN)在神经退行性疾病发病机制提供线索。方法以小鼠海马神经元来源的HT22细胞系为研究对象,采用细胞免疫荧光法和Western blot法检测PGRN在HT22细胞中的表达情况。结果 PGRN在HT22细胞胞浆和突起中表达,在细胞核周围表达尤为明显,并且PGRN可与神经元标记物βIII-tubulin和Dcx在HT22细胞中共表达。结论 HT22细胞高度表达PGRN,为PGRN在学习记忆中的作用研究提供了基础。
Objective To investigate the expression of progranulin (PGRN) in HT22 mouse hippocampal cells. This study will provide a clue for the further study of PGRN in the pathogenesis of neurodegenerative diseases. Methods HT22 cell line de- rived from mouse hippocampal neurons was cultured. Immunohistofluorescence and Western-blot methods were used to detect the ex- pression of PGRN. Results PGRN was expressed in cytoplasm and processes of HT22 cells, especially in the nucleus. PGRN can co-localize with neuronal markers Doublecortin (Dcx). Conclusion PGRN was highly expressed in HT22 cells.which provides a basis for the study of the role of PGRN in learning and memory.
