摘要 建立了一套适用于LC-MALDI系统的标记定量蛋白质组学分析技术,主要包括蛋白提取、溶液内酶解、肽段标记、液相分离、质谱检测、生物信息学分析等步骤。将该方法与传统的双向凝胶电泳技术进行了比较与评价,该方法的实验结果可靠、灵敏度高、实验周期短。为今后的蛋白质组学研究工作提供了有效的参考与借鉴。 This article sets up a set of quantitative proteomics analysis technology suitable for LC-MALDI system,including protein extraction,solution hydrolases,peptides labeling,liquid phase separation,massspectrum detection,bioinformatics analysis,etc.At the same time,this article compares it with twodimensional electrophoresis protocol and presents an assessment which has advantages of reliable experimental results,high sensibility and short test period.It provides an effective reference for proteomics research.
