摘要
目的:观察自拟扶正消癌Ⅰ号方对balbc小鼠移植瘤血管内皮生长因子(VEGF)表达的作用,探讨该方对多种肿瘤的作用机制。方法:采用balbc小鼠,脾下注射法建立鼠大肠癌细胞株肝转移的动物模型,随机分为4组,每组12只:1)阴性对照生理盐水组;2)氟尿嘧啶组;3)扶正消癌I号方中药组;4)扶正消癌I号中药+氟尿嘧啶组。观察生活质量,抑瘤率,免疫组化方法检测VEGF蛋白表达的情况。结果:扶正消癌Ⅰ号方+氟尿嘧啶组、消癌Ⅰ方组小鼠的生活质量较其他2组好,阴性对照组瘤体最大,瘤体中药组其次,联合用药组体积最小,氟尿嘧啶组、中药组及联合用药组均能下调VEGF蛋白的表达。结论:扶正消癌Ⅰ号方对鼠大肠癌细胞株肝转移小鼠抑制新生血管的作用机制可能是通过下调VEGF的表达,抑制肿瘤微血管生成,可能是该方抗肿瘤的机制之一。
Objective:To investigate the effect of Fuzheng Xiaoai I decoction on VEGF expression of mice with liver metastasis of colon cancer,and discuss its possible mechanism.Methods:The tumor models were established by injecting human carcinoma of CT26 cells under the balbc mice.Balbc mice with liver cancer cells are randomized divided into 4 groups (12 in each group):1) Control group;2)5-Fu group;3)Fuzheng Xiaoai I group;4)Fuzheng xiaoai I +5-Fu group.The life quality of mice,tumor inhib-iting rates,and the expression of VEGF in tumor tissue were measured.Results:The life quality of group 4 and 3 are better than that of the other groups.Besides,the tumor size of group 4 was the smallest,and that of the control group was the biggest.The levels of VEGF in all groups were decreased.Conclusion:Fuzheng Xiaoai I could inhibit the angiogenesis of tumor possibly by down regulating VEGF and obstructing the formation of tumor vessels.
