黄芩苷对大鼠牙周炎牙龈组织中TNF-αmRNA和IL-6mRNA表达的影响

Effects of baicalin on the expressions of TNF-αmRNA and IL-6mRNA in gingiva tissues of rat with periodontitis

目的观察黄芩苷对牙周炎大鼠牙龈组织中肿瘤坏死因子-α(TNF-α)mRNA、白细胞介素-6(IL-6)mRNA表达的影响,探讨黄芩苷治疗牙周炎的治疗机制。方法选用50只8周龄SD大鼠,随机分成5组,每组10只,A组为正常对照组,B组为牙周炎组,C1、C2、C3组为黄芩苷治疗组,B,C1,C2,C3组建立牙周炎模型,治疗1周后处死,采用RT-PCR方法检测TNF-αmRNA、IL-6mRNA在5组大鼠牙龈组织中的表达水平。结果牙周炎组大鼠牙龈组织中TNF-αmRNA、IL-6mRNA表达强度显著高于正常对照组(P<0.05),黄芩苷各治疗组大鼠牙龈组织中TNF-αmRNA、IL-6mRNA表达强度均明显低于牙周炎组(P<0.05)。结论黄芩苷可以降低牙周炎大鼠牙龈组织中TNF-αmRNA、IL-6mRNA的表达含量,并能改善大鼠牙周组织状况。

Objective To observe the effects of baicalin on the expressions of tumor necrosis factor alpha （ TNF-α） mRNA and interleukin-6 （IL-6） mRNA in gingiva tissues of rat with periodontitis in order to explore the action mechanism of baicalin in treatment of periodontitis.Methods Fifty SD rats （ 8-week age ） were randomly divided into 5 groups：blank control group （group A）, periodontitis group （group B）, baicalin treatment groups including group C1,group C2,group C3, with 10 rats in each group.The animal models with periodontitis were established in group B,group C1,group C2 and group C3.All the rats were sacrificed after 7-day treatment,and the expression levels of TNF-αmRNA and IL-6 mRNA were detected by reverse transcription polymerase chain reaction （ RT-PCR） technique.Results The expression levels of TNF-αmRNA and IL-6 mRNA in gingiva tissues of rats in group B were significantly higher than those in group A （ P 〈0.05）,moreover, the expression levels of TNF-αmRNA and IL-6 mRNA in group C1,group C2 and group C3 were significantly lower than those in group B （ P 〈0.05）.Conclusion The baicalin can decrease the expression levels of TNF-αmRNA and IL-6 mRNA in gingiva tissues of rat with periodontitis, and can also improve the status of periodontal tissues.
