阿里红提取物对Aβ_(25-35)致痴呆动物模型学习记忆障碍和海马乙酰胆碱酯酶活性的影响 被引量:4

摘要 目的观察阿里红提取物对Aβ_(25-35)诱导老年性痴呆大鼠的学习记忆力和海马乙酰胆碱酯酶活性的影响。方法采用侧脑室注射凝聚态Aβ_(25-35)蛋白诱导老年性痴呆(Alzheimer’s disease,AD)的大鼠模型。取SD大鼠60只,随机分为对照组、模型组(11.5μmol/L Aβ_(25-35))、阿里红提取物低剂量(11.5μmol/L Aβ_(25-35)+0.25 mg/kg阿里红乙酸乙酯部位)组、阿里红提取物中剂量(11.5μmol/LAβ_(25-35)+0.5 mg/kg阿里红乙酸乙酯部位)组、阿里红提取物高剂量(11.5μmol/L Aβ_(25-35)+1 mg/kg阿里红乙酸乙酯部位)组5组。对照组及模型组动物每天固定时间分别给予5μL的生理盐水和Aβ_(25-35),阿里红提取物各剂量组动物分别给予相应剂量的药物。SD大鼠连续给药10 d后,用Morris水迷宫方法对动物进行空间记忆能力测试;采用HE染色法观察各组大鼠神经元病理形态的改变;测定受试动物大脑海马组织乙酰胆碱酯酶(ACh E)含量。结果与模型组比较,阿里红提取物高、中、低剂量组大鼠逃逸潜伏期明显缩短,目标象限停留时间明显延长,穿越平台次数增加,差异有统计学意义(P<0.05);阿里红提取物高、中、低剂量组大鼠海马神经细胞数量明显增多,ACh E活性明显降低。结论阿里红提取物乙酸乙酯部位能改善Aβ_(25-35)诱导所致AD大鼠学习记忆能力障碍,亦能降低脑内乙酰胆碱酯酶活性。 Objective To observe the effects of Fomes officinalis extract on ability of memory and acetylcholinesterase activity in AD rats induced by Aβ_(25-35). Methods All groups except the control were injected Aβ_(25-35) into the lateral cerebral ventricle. 60 rats were randomly divided into 6 groups:the control group,the model group and 3experimental drug treated groups. Both control and model group were treated with appropriate dose of saline,3 experimental drug treated groups were treated with appropriate dose of Fomes officinalis ethyl acetate at fixed time every day. After administration for 10 days,the ability of memory of rats was tested by the water maze method and the content of ACh E was measured by Acetylcholinesterase Reagant Kit; Pathological changes of neurons in each group were observed with HE staining. Results Compared 3 experimental drug treated groups with the model group,the escape latency was significantly shortened,the residence time of target quadrant were significantly prolonged,the times of passing through the platform were significantly increased( P〈0 05);The number of nerve cells in hippocampus increased obviously,the activity of ACh E of 3 experimental groups was obviously lower than that of model group( P〈0 05). Conclusion Fomes officinalis extract of ethylacetate can improve the studying memory of theAD rats induced by Aβ_(25-35) and can also reduce the activity of ACh E.
