摘要
目的探讨丹红注射液(丹参和红花)对心梗(MI)模型大鼠缺血心肌血管新生的影响及其作用机制。方法利用Sprague-Dawley(SD)大鼠左前降支冠状动脉结扎法造成急性心梗模型,实验分为假手术组,模型组,丹红注射液低、中、高剂量组(0.5、1、2 m L/kg),静脉给药7 d后处死,检测心梗面积,心肌损伤血清学指标,微血管面密度,血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)、基质细胞衍生因子-l(stromal cell-derived factor-1,SDF-l)及其受体CXCR4的表达。结果丹红注射液可显著减小大鼠心肌梗死面积,降低LDH、CK-MB、c Tn I血清水平,增加微血管面密度,上调VEGF、SDF-l、CXCR4的表达。结论丹红注射液具有缺血心肌的保护作用,其机制可能与VEGF的高表达及SDF-l/CXCR4生物轴有关。
AIM To investigate the neovascular effect of Danhong Injection( Salviae miltiorrhizae Radix et Rhizoma and Carthami Flos) on myocardial infarction( MI) rats with myocardial ischemia and its mechanism of action. METHODS Sprague-Dawley( SD) rats with acute MI induced by ligation of left anterior descending coronary artery were randomly divided into five groups( sham-operated group,model group,0. 5 m L / kg,1 m L / kg and 2 m L / kg Danhong Injection groups). After intravenous injection for seven days,the rats were sacrificed. Then the infarct size,serum markers of myocardial damage,microvessel density,and expressions of vascular endothelial growth factor( VEGF),stromal cell-derived factor-1( SDF-l) and its receptor CXCR4 were determined. RESULTS Danhong Injection significantly decreased the infarct size, serum levels of lactate dehydrogenase( LDH),creatine kinase-MB( CK-MB) and cardiac troponin I( c Tn I),increased the microvascular density,and upregulated the expressions of VEGF,SDF-l and CXCR4 in rats. CONCLUSION Danhong Injection has cardioprotective effect,which may be related to the high expression of VEGF and SDF-l / CXCR4 biological axis.
