摘要
目的建立RP-HPLC法同时测定白头翁汤(白头翁、黄连、黄柏、秦皮)中8种成分的含有量。方法白头翁汤水提液的分析采用Agilent ZOBAX XDB-C18色谱柱(250 mm×4.6 mm,5μm);以0.1%磷酸(p H=3.7)-乙腈为流动相,梯度洗脱;体积流量0.7 m L/min;检测波长315 nm(秦皮甲素、秦皮乙素)、284 nm(黄柏碱)、345 nm(表小檗碱、盐酸小檗碱、巴马汀、黄连碱)、210 nm(白头翁皂苷B_4)。结果秦皮甲素、黄柏碱、秦皮乙素、表小檗碱、巴马汀、黄连碱、盐酸小檗碱、白头翁皂苷B_4分别在0.5~160、64~640、0.75~150、0.5~100、1.35~550、0.12~140、0.7~140、0.5~170 ng/m L范围内线性关系良好,平均回收率均大于96%,RSD均在2.5%以下。结论该方法准确灵敏,可用于白头翁汤的质量控制。
AIM To establish an RP-HPLC( reversed phase high performance liquid chromatography) method for the simultaneous determination of eight constituents in Baitouweng Decoction( Pulsatillae Radix,Coptidis Rhizoma,Phellodendri Chinensis Cortex and Fraxini Cortex). METHODS The analysis of aqueous extract of Baitouweng Decoction was carried out on an Agilent ZOBAX XDB-C18column( 250 mm × 4. 6 mm,5. 0 μm),with the mobile phase comprising of 0. 1% phosphoric acid( p H = 3. 7)-acetonitrile flowed at 0. 7 m L / min in a gradient elution manner,and the detection wavelengths were set at 315 nm for aesculin and aesculetin,284 nm for phellodendrine,345 nm for epiberberinem,berberine hydrochloride,palmatine and coptisine,and 210 nm for anemoside B4. RESULTS Aesculin,phellodendrine,aesculetin,epiberberine,palmatine,coptisine,berberine hydrochloride and anemoside B4 showed good linear relationships within the ranges of 0. 5- 160 ng / m L,64- 640 ng / m L,0. 75- 150 ng / m L,0. 5- 100 ng / m L,1. 35- 550 ng / m L,0. 12- 140 ng / m L,0. 7- 140 ng / m L and 0. 5-170 ng / m L,respectively. All of the average recoveries were over 96% with the RSDs of less than 2. 5%. CONCLUSION This accurate and sensitive method can be used for the quality control of Baitouweng Decoction.
