RIP3蛋白在重症急性胰腺炎小鼠胰腺组织中表达 被引量:4

摘要 目的观察胰腺组织受体相互作用蛋白3(RIP3)的表达变化,初步探讨RIP3与胰腺细胞坏死的关系。方法建立重症急性胰腺炎小鼠模型,HE染色观察胰腺组织形态,应用免疫组化法测定RIP3蛋白的表达。结果对照组及研究组胰腺组织结构清晰,细胞形态正常,无出血坏死。研究组小鼠6 h呈现水肿胰腺炎改变;12、24 h胰腺小叶结构紊乱,明显充血,腺泡细胞坏死,坏死区周围炎细胞侵润。免疫组化显示,对照组RIP3胰腺组织阴性表达;研究组RIP3表达随着时间推移逐渐增强,12、24 h研究组RIP3蛋白表达较对照组显著升高;免疫组化平均光密度值示3 h与对照组比较,差异无统计学意义(P>0.05);6、12、24 h与对照组比较,差异有统计学意义(P<0.05)。结论 RIP3蛋白可能在胰腺细胞坏死过程中具有重要作用。 Objective To explore the expression changes of receptor interacting protein 3( RIP3) in pancreatic tissue of severe acute pancreatitis mice and the relationship between RIP3 and necrosis of pancreatic cells preliminary. Methods To establish model of severe acute pancreatitis in mice. Pancreatic tissues were stained by HE and observed under microscope. The expression of RIP3 protein in pancreatic tissue was determined by immunohistochemistry. Results In the control group and study group,the pancreatic organizational structure was clear,cell morphology was normal without bleeding necrosis. In the study group,the change of edema pancreatitis appeared in mice after 6 hours,there was disordered pancreatic lobule structure,marked hyperemia,acinar cell necrosis and inflammatory cells invasion from the area around necrosis. Immunohistochemistry showed that RIP3 protein expression was negative in the control group. In the study group,RIP3 expression increased gradually as time increased,The RIP3 protein expression in study group after12 hours and 24 hours were significantly higher than the control group( P〈0. 05). The immunohistochemical average optical density value after 3 hours had no statistical significance( P〉0. 05) which compared with the control group,however,there was statistical significance after 6 hours,12 hours and 24 hours( P〈0. 05). Conclusion It is speculated that RIP3 protein in the process of pancreatic cell necrosis plays an important role.
