摘要
目的研究重组人脑利钠肽联合曲美他嗪对异丙肾上腺素诱发心力衰竭大鼠心肌重构的影响。方法采用连续腹腔注射异丙肾上腺素制备Sprangue-dawly(SD)大鼠慢性心衰模型。存活者随机分为心衰组治疗组(n=5)(给予重组人脑利钠肽22.5μg/kg/d皮下注射,曲美他嗪片10 mg/kg/d灌胃)及对照组(n=5)(给予同等剂量生理盐水皮下注射及灌胃)。干预1个月后取大鼠心脏标本观察心肌细胞形态学变化、左心室重量(left ventricular mass,LVM)及左心室重量指数(left ventricular mass index,LVMI)及心肌组织内胶原蛋白含量检测。结果心肌组织的HE染色显示:对照组心肌纤维明显肥大,而治疗组心肌纤维较对照组明显纤细。微观结构:治疗组心肌损伤程度较对照组明显减轻。心肌组织重量方面:治疗组较对照组LVM、LVMI差异有统计学意义(P<0.05)。胶原蛋白含量方面:治疗组较对照组胶原蛋白含量更低,组间差异均有统计学意义(P<0.05)。结论重组人脑利钠肽联合曲美他嗪治疗心力衰竭疗效显著。
Objective The effects of rhBNP and trimetazidine in induced of isoproterenol heart failure rats. Methods Ten male SD rats were randomly divided into control group and model group. Rats in model group were injected subcutaneously with isoproterenol. Four weeks later, the rats in model group were randomly divided into two groups: rhBNP and trimetazidine treatment group(rhBNP 22.5 /~g/kg, once daily, Subcuta- neous injection; trimetazidinel0 mg/kg, once daily, gavage; n = 5), sham operation group(only received an e- qual volume of normal saline, n = 5). The indexes of LV remodeling and electron microscopy were examined afer 4 weeks. Results Compared with myocardial biopsy of model group, myocardial cell of SHR control group showed significant injury; compared with model group, electron microscopy showed that the ultrastructure of myocardium in SHR control group were in serious disorder, and the improvement even more significant in in model group ; there was no significant difference among each group in terms of BW(P 〉0.05), the LVW, LV- MI, collagen content in model control group were significantly higher than those in control group( P 〈 0.05), after drug treatment, LVW, LVMI, collagen content in common model group and control group were significant- ly reduced(P 〈 0.05). Conclusion The rhBNP and trimetazidine can reduce myocardial cell injury in rat with HF, and signicantly inbibjt the development of LV remode ling and improve cardiac function.
