产丝素蛋白酶菌株发酵条件的优化

摘要 采用单因素法,研究了产丝素蛋白酶菌株在发酵过程中,培养温度,培养基初始p H值,培养基碳源、氮源、金属离子等因素对产丝素蛋白酶菌株的发酵影响,最后通过对发酵产生的产丝素蛋白酶酶活及丝素蛋白降解的影响分析,来初步确定最佳发酵条件。研究结果表明:培养温度30℃,培养基初始p H值8.0,培养基中添加丝素,以玉米粉作为碳源,KNO3为氮源,以Mg2+作为金属离子,在该条件下发酵所得丝素蛋白酶活性最高,降解程度最大。 By single test, choosing culture temperature,initial pH value, carbon sources, nitrogen sources, metal ion as the conditions, effect of the fermentation for fibroinase strain was studied. Then through the effect on the fibroinase activity and fibroin loss rate,the best conditions for fermentation was chose.The results showed that best fermentation conditions for fibroinase strain was as followed:the culture temperature 30 ℃,initial pH 8.0, corn powder as the carbon sources, KNO3 as the nitrogen sources, Mg^2+ as the metal ion, and add some fibron.Under the condition,we can get the best fibroinase activity and fibroin loss rate.
