摘要
目的:探讨一种新型卟啉类光敏剂DTP对人食管癌细胞EC109的光动力学治疗(PDT)作用及机制。方法:采用MTT法检测DTP-PDT对食管癌细胞EC109的杀伤作用;酶标仪检测EC109细胞内DTP吸收量;激光共聚焦显微镜观察DTP在肿瘤细胞内定位;流式细胞术检测DTP-PDT后细胞死亡方式。结果:单纯药物孵育对EC109细胞生长无任何影响,但DTP-PDT显示出对食管癌细胞EC109较强的杀伤作用;EC109细胞内DTP吸收呈显著的剂量依赖性;DTP分布于EC109细胞溶酶体内;DTP-PDT后细胞死亡方式主要为凋亡。结论:新型光敏剂DTP光动力学治疗能够有效杀伤人食管癌EC109细胞,主要方式为通过损伤溶酶体间接启动细胞凋亡。
Objective:To investigate the photodynamic therapy(PDT)effects and the mechanisms of a novel porphyrin-based photosensitizer PDT on human esophageal carcinoma cell line EC109.Methods:MTT assay was used to investigate the DTP-PDT mediated cytotoxicity on esophageal carcinoma cell line EC109;Intracellular DTP uptake was determined with luminescence spectrometer and intracellular DTP distribution site was observed by laser scanning confocal microscopy(LSCM);Flow cytometry was used to detect the cell death mode after DTP-PDT treatment.Results:DTP alone showed no any influence on the growth of EC109 cells,however,DTP-PDT treatment exhibited significant cytotoxic effects on EC109cells;Intracellular DTP accumulation presented an obviously dose-dependent trend;DTP localized in the lysosomes of EC109cells;Apoptosis was the main death mode of EC109 cells after DTP-PDT treatment.Conclusion:The novel photosensitizer DTP mediated photodynamic therapy could effectively kill human esophageal carcinoma cell line EC109,which was achieved through the initiation of indirect cell apoptosis induced by lysosomal destruction following DTP-PDT treatment.
