HPLC同时测定柿叶提取物中槲皮素-7-O-β-D-鼠李糖苷和染料木素含量的方法研究

Simultaneous Determination of Quercetin-7-O-β-D-rhamnoside and Genistein in Extract of Persimmon Leaves by HPLC

目的：建立柿叶提取物中槲皮素-7-O-β-D-鼠李糖苷和染料木素的HPLC同时测定方法。方法：采用HPLC法,Phenomenex-C18（250mm×4.6mm,5μm）,甲醇-0.02%磷酸水为流动相梯度洗脱,检测波长：368nm,流速：1mL/min;柱温30℃。结果：槲皮素-7-O-β-D-鼠李糖苷和染料木素分别在7.44-297.4μg/mL范围和1.10-109.8μg/mL范围内线性关系良好,平均回收率分别为99.20%和98.77%。结论：该方法准确、可靠,重复性好,可用于柿叶提取物的质量评价与控制标准,为柿叶功能性产品的研究开发奠定基础。

Objective：To establish a HPLC method for simultaneous determination of the content of quercetin-7-O-β-Drhamnoside and genistein in extract of persimmon leaves.Methods：The HPLC analysis was carried out on Phenomenex-C18column（250mm×4.6mm,5μm）with methanol-0.02% phosphoric acid solution as the mobile phase for gradient elution at a flow rate of 1mL/min.The detection wavelength was 368 nm and the column temperature was 30℃.Results：Good linear relationship were found in the range of 7.44~297.4μg/mL for quercetin-7-O-β-D-rhamnoside,1.10~109.8μg/mL for genistein.Their average recoveries were 99.20% and 98.77% respectively.Conclusion：The method is accurate,reliable,and can be used for the quality control and evaluation of extract of persimmon leaves.It lays the foundation for the research and development of persimmon leaf functional products.
