摘要
目的:观察白藜芦醇(Res)对人胰腺癌Miapaca-2(PC-2)细胞构建的胰腺癌裸鼠皮下移植瘤生长的抑制作用,并探讨其作用机制。方法:建立胰腺癌裸鼠移植瘤模型并随机分组,分别给予3种不同浓度的白藜芦醇干预后处死胰腺癌裸鼠,取出移植瘤,观察其体积、重量的变化情况;HE染色法观察移植瘤组织病理学变化;采用实时定量聚合酶链反应(RT-PCR)和Western bolt法测定移植瘤组织Bcl-2、Bax、p53蛋白和mRNA表达水平。结果:Res组移植瘤的体积、重量明显低于阴性对照组,随着浓度增加变化越明显(P<0.01);3个浓度组白藜芦醇作用于胰腺癌裸鼠后,移植瘤组织中Bcl-2、Bax、p53蛋白和mRNA表达水平较阴性对照组差异具有统计学意义(P<0.01),且随着浓度增加表达水平的差异越明显。结论:白藜芦醇可能通过调节Bcl-2、Bax、p53的表达来抑制胰腺癌移植瘤的生长。
Objective:To investigate the antitumor effect of Resveratrol on pancreatic cancer Miapaca-2 cell xenograft on athymic.Methods:The pancreatic cancer Miapaca-2 cell xenograft model in nude mice was established and randomly divided into the blank group,the Res group and the saline negative control group.After intervention with Res,the mice was killed.The weight and volume of tumor were recorded.The pathological change of tumor tissue in each group was observed under microscope with HE staining.Real-time quantitative RT-polymerase chain reaction and western bolt assay were used to confirmed the expression of Bcl-2 and Bax、p53 in tumor tissue at mRNA and protein lever.Results:The weight and volume of tumor in Res group were less than those in the negative control group(P〈0.01).RT-PCR and Western blot assay indicated Resveratrol of three different concentrations regulated the protein and mRNA expression of Bcl-2 and Bax、p53 in tumor tissue.At the same time,the expression levels gradually changed as the concentration increased,the difference was significant(P〈0.01).Conclusion:Resveratrol inhibited the growth of pancreatic cancer Miapaca-2 cells xenograft.The effect may be related to the regulating of Bcl-2 and Bax、p53at mRNA and protein lever.
