PTEN、VEGF在人脑星形细胞瘤的表达及意义

摘要 目的探讨PTEN及VEGF与脑星形细胞瘤发生、发展的关系,寻找抑制脑星形细胞瘤生长和侵袭的有效方法。方法方便选取2012年11月—2015年11月该院收治的49例星形细胞瘤采用免疫组化SP法检测了49例脑星形细胞瘤及正常脑组织中PTEN基因蛋白及血管内皮细胞生长因子的表达情况,并探讨了PTEN与VEGF的相互关系。结果脑星形细胞瘤组织中PTEN蛋白表达率显著低于正常脑组织(P〈0.05),且瘤组织分化程度越低,PTEN阳性率越低,差异具有统计学意义(P〈0.05);VEGF蛋白表达在正常脑组织为阴性,且在Ⅰ~Ⅱ级与Ⅲ~Ⅳ级星形细胞瘤(r=-0.342,P〈0.05)、瘤组织与正常组织相比均差异有统计学意义(P〈0.01);PTEN蛋白的表达和VEGF蛋白表达之间呈负相关关系(P〈0.05)。结论脑星形细胞瘤组织中PTEN蛋白表达的减少或缺失,均可导致VEGF的表达上升,进而生成肿瘤血管,最终为脑星形细胞瘤的生长和浸润提供重要的基础。 Objective PTEN and VEGF and astrocytoma, development of relations, to find an effective method of brain astrocytoma growth and invasion inhibition. Methods Convenient select November 2012 to November 2015 in the hospital were 49 cases of astrocytoma in 49 cases of astrocytoma and normal brain tissue PTEN protein and vascular endothelial cell growth factor expression was detected by immunohistochemical staining(SP method), and to investigate the relationship between PTEN and VEGF. Results Astrocytoma of PTEN protein expression was significantly lower than in normal brain tissue(P〈0.05), and the lower the degree of differentiation of tumor tissue, the lower the rate of PTEN, a statistically significant difference(P〈0.05); VEGF protein expression in normal brain tissue was negative, and in grade Ⅰ ~ Ⅱ and Ⅲ ~ Ⅳgrade astrocytomas(P〈0.05), tumor tissue and normal tissue(P 0.01) were significantly different compared;. PTEN protein negative correlation(r=-0.342,P〈0.05) between the expression and protein expression of VEGF. Conclusion Astrocytoma of PTEN protein expression is reduced or absent, can lead to increased expression of VEGF, thereby generating tumor blood vessels, and ultimately provide an important basis for the growth of brain astrocytoma and infiltration.
