摘要
为研究甘蓝SLR1启动子在异源植物中的表达特点,将GUS报告基因在1.5kb长的SLR1启动子驱动下,通过农杆菌介导法分别导入烟草和芥菜.转基因植株经PCR鉴定显示GUS报告基因已经整合到烟草和芥菜基因组中.转基因植株开花后花器官组织的GUS化学染色分析结果显示SLR1启动子主要在烟草和芥菜开花前2天至开花后的柱头中大量表达,部分转基因烟草植株开花后的花冠,转基因芥菜植株花萼、花瓣及花丝中也有明显的表达,但在两种转基因植株花粉中均未检测到GUS表达活性,显示甘蓝SLR1启动子在异源植物中不是一个严谨的柱头特异启动子.
To explore Brassica oleracea SLR1 gene promoter expression pattern in heterologous plant,the1.5-kb SLR1 promoter has ligated upstream of the GUS reporter gene,and has been introduced into tobacco and tuber mustard through Agrobacterium mediated transformation respectively.PCR results indicate the GUS reporter gene has been integrated into the genome of tobacco and tuber mustard.Based on the histochemical localization of GUS activity of the flowers organs and tissues,all transgenic plants show robust GUS activity in the stigma at 2days prior to flower opening,reaches a maximum in opening flowers,and persists until flower senescence.In addition,GUS activity has also been detected in the corolla of a few transgenic tobacco plants,the calyxes,petals and filaments of a few transgenic tuber mustard plants.However,no GUS activity has been detected in the pollen of the tobacco and tuber mustard transgenic plants,All these results show the Brassica oleracea SLR1 gene promoter is not a rigorous stigma-specific promoter expressed in heterologous plants.
