摘要
目的观察芪珀生脉颗粒对房颤大鼠模型cAMP、cGMP、cAMP/cGMP的影响。方法采用乙酰胆碱-氯化钙混合液通过尾静脉给药建立房颤大鼠模型。根据预实验房颤造模死亡情况估计,从90只SD大鼠中随机抽取8只作为正常组,其余大鼠分为模型组、稳心颗粒组、维拉帕米组、芪珀生脉颗粒大剂量组(大剂量组)、芪珀生脉颗粒中剂量组(中剂量组)、芪珀生脉颗粒小剂量组(小剂量组)。治疗4周后,测定各组大鼠血浆cAMP、cGMP,计算cAMP/cGMP并进行统计学分析。结果各组间cAMP水平差异无统计学意义(P>0.05);模型组cGMP水平低于除小剂量外的各组(P<0.05),其中与正常组、维拉帕米组、大剂量组间差异有统计学意义(P<0.01);各组cGMP水平均高于小剂量组(P<0.05),其中正常组、维拉帕米组、大剂量组间差异有统计学意义(P<0.01);模型组cAMP/cGMP水平高于正常组、维拉帕米组、大剂量组,组间差异有统计学意义(P<0.05)。结论芪珀生脉颗粒能够有效升高房颤大鼠cGMP,降低cAMP/cGMP。
Objective To observe the effect of Qipo Shengmai Granules on cAMP, cGMP and cAMP/cGMP in model rats with atrial fibrillation(AF). Methods ACh-CaCl2 mixed liquid was injected through the caudal vein of rats to establish the model with atrial fibrillation. Estimated according to the condition of death during the pre-experiment of model establishing, 8 rats were selected randomly as the normal group among 90 SD rats, others as the model group, Wenxin Granules group, verpamil group, Qipo Shengmai Granules high-dose group (high-dose group), Qipo Shengmai Granules middle-dose group (middle-dose group), Qipo Shengmai Granules low-dose group ( low-dose group) respectively. The level of cAMP, cGMP and cAMP/cGMP in plasma of AF rats were measured for statistical analysis 4 weeks later. Results There were no statistical differences in cAMP level among all the groups ( P 〉 0.05). The level of cGMP in model group was lower than other groups, except the low-dose group (P 〈 0. 05 ), of them, the differences between the model group with the normal group, verpamil group, and high-dose group were statistically significant (P 〈 0. 01 ) ; and cGMP levels of all groups was higher than that of low-dose group, the differences between the model group with the normal group, verpamil group, and high-dose group were statistically significant(P 〈0. 01 ). The level of cAMP/cGMP in model group was higher than that of the normal group, verpamil group and large-dose group, the differences were statistically significant (P 〈 0.05 ). Conclusion Qipo Shengmai Granules could enhance the level of cGMP and depress the level of cAMP/cGMP in AF rats effectively.
