天麻钩藤饮不同分离部位对偏头痛模型大鼠的影响

摘要 目的:观察天麻钩藤饮大孔树脂不同分离部位对偏头痛模型大鼠的影响。方法:按随机数字表法将70只SD大鼠随机分为阳性对照组(西比灵组)、正常对照组、模型对照组、天麻钩藤饮(TMGTY)组、天麻钩藤饮30%part组(以下简称30%part组)、天麻钩藤饮60%part组(以下简称60%part组)、天麻钩藤饮90%part组(以下简称90%part组)共7组。采用皮下注射硝酸甘油注射液复制偏头痛大鼠模型;采用时间分段计数法持续观察造模后大鼠耳红出现和消失时间、挠头等行为学的变化情况;采用酶免法测定各组大鼠血清5-羟色胺(5-HT)和肾上腺素(E)含量。结果:正常对照组未出现耳红症状,其余各组均出现耳红症状(P〈0.01)。除正常对照组外,各组耳红出现时间整体比较,差异无统计学意义(P〉0.05)。与模型对照组比较,TMGTY组、60%part组的耳红消失时间均缩短(P〈0.05);30%part组、90%part组大鼠耳红消失时间虽有缩短趋势,但差异无统计学意义(P〉0.05)。模型对照组在皮下注射10 mg/kg硝酸甘油注射液后30-150 min的挠头次数较正常对照组增加(P〈0.01)。与模型对照组比较,60%part组在0-150 min内不同时间段的挠头次数均减少(P〈0.05,P〈0.01);TMGTY组仅在30-60 min时间段挠头次数减少(P〈0.05);30%part组和90%part组仅在120-150 min时间段时间段挠头次数减少(P〈0.05)。与正常对照组比较,模型组对照组的血清5-HT含量降低(P〈0.01),血清E含量升高(P〈0.05)。与模型对照组比较,TMGTY组、60%part组和90%part组的血清5-HT水平均升高(P〈0.01),30%part组的血清5-HT水平有升高趋势,但差异无统计学意义(P〉0.05);TMGTY组、30%part组、60%part组和90%part组的血清E水平均下降(P〈0.05,P〈0.01)。结论:天麻钩藤饮60%乙醇洗脱部位能有效改善模型大鼠耳红、挠头等症状,调节模型大鼠的血清5-HT、E含量。 Objective: To observe macroporous resin fraction of Tianma Gouteng Yin on migraine model rats, to provide reference for selecting effective parts of Tianma Gouteng tang anti-migraine. Methods: Seventy rats were divided into seven groups: positive control group, normal control group, model control group, Tianma Gouteng tang(TMGTY)group, TMGTY 30%part, 60%part, 90%part group. Copied migraine models by injecting subcutaneously nitroglycerin injection, observed constantly appearance and disappearance time of red ears, as well as ethology changes including scratching head after modeling by time slicing counting method, detected levels of 5-hydroxytryptamine(5-HT) and epinephrine(E)in serum of every group by ELISA method. Results. There was no symptom of red ears in normal control group, other groups appeared symptom of red ears(P〈 0.01). Except normal control group, comparing appearance time of red ears in each group, there was no significant difference(P〉 0.05). Comparing with model control group, disappearance time of red ears in TMGTY group and TMGTY 60 % part group was reduced(P〈0.05); although disappearance time of red ears in 30%part and 90%part group had reduction tendency, there was no statistical significance (P 〉 0.05). The phenomenon of scratching head appeared in model control group after 30-150 min of injecting 10 mg/kg nitroglycerin injection subcutaneously(P〈 0.01 compared with before). Comparing with model control group, the number of scratching head on different time period during 0-150 min in 60% part group were all reduced(P 〈 0.05, P〈 0.01); the number of scratching head were reduced only during 30-60 min in TMGTYgroup(P 〈 0.05); the number of scratching head were reduced only during 120-150min in 30% part and 90% part group(P 〈 0.05). Comparing with normal control group, levels of 5-HT in serum of model control group were reduced(P〈 0.01), levels of E in serum were increased(P〈 0.05). Comparing with model control group, levels of 5-HT in serum of TMGTY group, 60% part group and 90% part group were all increased (P 〈0.01); levels of 5-HT in serum of 30%part group had increasing tendency(P〉0.05) . Levels of E in serum of TMGTY group 30% part group, 60% part group and 90% part group were all reduced (P 〈 0.05, P 〈0.01). Conclusion. TMGTY 60% part can improve red ears, scratching head and other syndromes effectively, regulate levels of 5-HT and E in rat.
