摘要
目的优化黄连解毒流浸膏制备工艺。方法首先进行提取方式(分煎与合煎)的比较,然后采用单因素试验考察提取溶剂、提取时间、提取温度、料液比、乙醇浓度对提取率的影响;选取提取时间、乙醇浓度及料液比为考察因素,采用响应面优化试验(Box-Behnken)优选最佳提取工艺。结果最佳提取工艺为:黄连和黄柏加11.8倍61%乙醇,86℃下提取123 min,黄芩和栀子加6倍60%乙醇,82℃下提取123 min。结论此方法操作简单、提取率高,黄连解毒流浸膏质量稳定,可用于后续黄连解毒颗粒的制备。
Objective To optimize the preparation process of Huanglian Jiedu Fluid extract. Methods First, the results of different extraction patterns(separated and mixed boiling) were compared. Then the single factor experiment was used to observe the effects of extraction solvent, extraction time, extraction temperature and solid-liquid ratio on extraction rate. The response surface optimization(Box-Behnken) test was used to screen the optimum extraction process, with extraction time, ethanol concentration and solid-liquid ratio as factors. Results The optimum process was as follows: Coptidis rhizome and Phellodendri chinensis cortex were extracted for 123 min at 86 ℃, with 11.8 times of 61 % ethanol as extraction solvent; Scutellariae radix and Gardeniae fructus were extracted for 123 min at 82 ℃, with 6 times of 60 % ethanol as extraction solvent. Conclusion This method is simple and of high extraction rate, and the quality of Huanglian Jiedu Fluid extract is stable, thus can be used for subsequent preparation of Huanglian Jiedu Granules.
