高通量测序分析红斑期酒渣鼻患者与健康人面部皮肤微生物的分布差异 被引量:2

The Difference of Facial Skin Microbiome Distribution Between Rosacea Patients and Healthy People through High-Throughput Sequencing

摘要 目的用16S rRNA基因序列分析技术来研究红斑期酒渣鼻患者和健康人面部皮肤微生物是否有差异,从而明确微生物是否是导致红斑期酒渣鼻的主要原因。方法用无菌敷料粘贴受试者左侧鼻翼及面颊的办法获取标本,进行DNA抽提,构建16S rRNA文库,然后将混合样本送至公司测序和鉴定微生物。结果 20个标本微生物丰度均合格,共测出746 631条16S rRNA序列进行下一步的分析,发现红斑期酒渣鼻组及健康组的大部分皮肤微生物均属于变形菌门、厚壁菌门、放线菌门、拟杆菌门、异常球菌-栖热菌门,其中属于变形菌门的沙雷氏菌、不动杆菌丰度最高,二组未发现有显著差异的微生物(P<0.01)。结论微生物不是导致红斑期酒渣鼻的主要病因。 Objective We study microbiome distribution of facial skin of the patients with erythematotelangiectatic rosacea and normal facial skin using 16S rRNA gene sequence analysis technology, and analyze whether there is difference between them. Thus, to explicit whether the microbiome is the main cause of erythematotelangiectatic rosacea. Methods We collect the samples of skin surface on left alar and cheer using the Tegaderm Film. Prior to DNA extraction, building 16S rRNA sequence libraries, then send the samples to company to identify microorganisms. Results The microbial abundance of the 20 samples are all qualified, and we analyzed 746 631 sequences for further phylogenetic analysis. We found that the two groups had the core microbiota, they were Proteobacteria, Firmicutes, Actinobacteria, Bactereoidetes, Cyanobacteria/Chloroplast, and the majority of these sequences were Proteobacteria phylum and predominantly genera Serratia and Acinetobacter. And we found no significant difference between microbes(P 〈0. 01 ). Discussion Microorganisms are not the main cause of erythematotelangiectatic rosacea.
