二甲双胍对细菌脂多糖诱导下腹腔巨噬细胞分泌IL-6的影响

摘要 目的探讨二甲双胍对细菌脂多糖(LPS)诱导下腹腔巨噬细胞分泌IL-6的影响。方法提取C57BL/6小鼠腹腔巨噬细胞并培养,采用MTT法检测不同浓度二甲双胍(1,5,10mmol/L)对腹腔巨噬细胞存活率的影响;采用ELISA法和免疫细胞化学法分别检测对照组、LPS诱导组、LPS诱导+不同浓度药物组腹腔巨噬细胞IL-6的分泌水平。结果 MTT法检测结果显示,在不同浓度二甲双胍作用下巨噬细胞存活率与对照组比较,差异无统计学意义(P=0.95)。与对照组比较,LPS诱导下腹腔巨噬细胞分泌的IL-6水平明显升高(P<0.01);与LPS诱导组比较,LPS诱导+二甲双胍组中IL-6水平降低,且IL-6水平下降程度与二甲双胍存在剂量依赖性关系(P<0.01)。结论二甲双胍能够抑制LPS诱导下腹腔巨噬细胞IL-6的分泌。 Objective To explore the effects of Metformin on the production of Interleukin-6 ( IL-6 ) in peritoneal macrophages of mice induced by LPS. Methods Isdating the peritoneal macrophages from C57BL/6 mice strain and then culure cells. MTT was used to detect the cell viability with various concentrations ( 1,5, 10mmol/L) of Metformin. ELISA and Immunocytochemistry method were used to measure the production of IL-6 in control group and LPS-induced group and various concentrations Metormin with LPS-induced groups. Results There was no statistically significant difference in cell viability between various concentrations Metformin groups and control group measured by MTT assay( P = 0. 95 ). The production of IL-6 was higher in LPS-induced group compared with control group(P 〈 0. 01 ). The production of IL-6 was lower in various concentrations Metformin with LPS-induced groups compared with LPS-induced group. There was a doesdependent relationship between degree of decrease of IL-6 and Metformin (P 〈 0. 01 ). Conclusion Metformin inhibits the release of IL-6 in LPS-induced peritoneal macrophages.
